「make sure + to不定詞」と「make sure + that節」は以下のように使い分けます。
①「make sure + to不定詞」は「必ず〜するようにしている」という意味を表すときに使用します。
I make sure to lock the door before I leave.
私は、出かける前に必ずドアに鍵をかけるようにしている。
②「make sure + that節」は「〜であることを確認する/〜であるか確認する」という意味を表すときに使用します。
I make sure that I have locked the door before I leave.
私は、出かける前にドアに鍵をかけたか確認する。

「 despite」の方が「in spite of」よりフォーマルな表現です。 両方とも「〜にも関わらず」という意味を持ち、一般的に口頭より書面で使用されます。尚、一般的に名詞（句）が続きます。
The game continued despite the heavy rain.
The game continued in spite of the heavy rain.
その試合は、激しい雨にも関わらず続いた。
名詞（句）の代わりに動名詞が使用されることもあります。
Despite achieving his goals, he wasn’t promoted.
In spite of achieving his goals, he wasn’t promoted.
彼は目標を達成したにも関わらず、昇進しなかった。
また、「despite」と「in spite of」の後にthat節を続ける場合、that節の前に「the fact」が必要になります。
He was late to work despite the fact that he woke up early.
He was late to work in spite of the fact that he woke up early.
彼は早く起きたにも関わらず、仕事に遅刻した。
因みに、「out of spite」は「腹いせに、悪意で」という意味になります。
He broke the car window out of spite.
彼は悪意で車の窓を割った。
